ABSTRACT

SARS-CoV-2, a newly discovered coronavirus, has been linked to the COVID-19 pandemic and is currently an important public health issue. Despite all the work done to date around the world, there is still no viable treatment for COVID-19. This study examined the most recent evidence on the efficacy and safety of several therapeutic options available including natural substances, synthetic drugs and vaccines in the treatment of COVID-19. Various natural compounds such as sarsapogenin, lycorine, biscoclaurine, vitamin B12, glycyrrhizic acid, riboflavin, resveratrol and kaempferol, various vaccines and drugs such as AZD1222, mRNA-1273, BNT162b2, Sputnik V, and remdesivir, lopinavir, favipiravir, darunavir, oseltamivir, and umifenovir, resp., have been discussed comprehensively. We attempted to provide exhaustive information regarding the various prospective therapeutic approaches available in order to assist researchers and physicians in treating COVID-19 patients.

ABSTRACT

Administrative restrictions on public mobility may impose significant well-being impacts on society. For instance, there has been a wide discussion on the psychological effects of lockdown policies during the pandemic. However, there is limited knowledge regarding the consequences of the policy transition and the uncertainty that arises from shifting from tight to loose regulations. The unexpected removal of Zero-COVID policies in China provides a special opportunity to study how policy uncertainty may affect mental health or subjective well-being. We use the Baidu Index data to test the effects of switching toward a "live-with-COVID” policy and the consequent rapid COVID spread on the frequency of well-being related search terms in China. Through a three-phase difference-in-differences approach with multiple years to check parallel sentiment trends, we find a substantial increase in fear and social dysfunction during the Transition Period when COVID-related restrictions were not fully lifted, as well as in the following Free-Spread Period. In comparison, the increase of anxiety mainly took place in the Transition Period. However, searches for anger and negative behaviors stayed relatively stable. Our results suggest that the removal of Zero-COVID policies, especially the Transition Period that generated policy uncertainty, significantly affected people's mental health.

ABSTRACT

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is a new type of coronavirus that has caused fatal infectious diseases and global spread. This novel coronavirus attacks target cells through the interaction of spike protein and angiotensin-converting enzyme II (ACE2), leading to different clinical symptoms. However, for a successful pregnancy, a well-established in-uterine environment includes a specific immune environment, and multi-interactions between specific cell types are prerequisites. The immune-related changes in patients infected with novel coronavirus could interfere with the immune microenvironment in the uterus, leading to fetal loss. We first reviewed the intrauterine environment in the normal development process and the possible pregnancy outcome in the infection state. Then, we summarized the immune response induced by SARS-CoV-2 in patients and analyzed the changes in ACE2 expression in the female reproductive system. Finally, the present observational evidence of infection in pregnant women was also reviewed.

ABSTRACT

The purpose of resource scheduling is to deal with all kinds of unexpected events that may occur in life, such as fire, traffic jam, earthquake and other emergencies, and the scheduling algorithm is one of the key factors affecting the intelligent scheduling system. In the traditional resource scheduling system, because of the slow decision-making, it is difficult to meet the needs of the actual situation, especially in the face of emergencies, the traditional resource scheduling methods have great disadvantages. In order to solve the above problems, this paper takes emergency resource scheduling, a prominent scheduling problem, as an example. Based on Vague set theory and adaptive grid particle swarm optimization algorithm, a multi-objective emergency resource scheduling model is constructed under different conditions. This model can not only integrate the advantages of Vague set theory in dealing with uncertain problems, but also retain the advantages of adaptive grid particle swarm optimization that can solve multi-objective optimization problems and can quickly converge. The research results show that compared with the traditional resource scheduling optimization algorithm, the emergency resource scheduling model has higher resolution accuracy, more reasonable resource allocation, higher efficiency and faster speed in dealing with emergency events than the traditional resource scheduling model. Compared with the conventional fuzzy theory emergency resource scheduling model, its handling speed has increased by more than 3.82 times.

ABSTRACT

In December 2019, coronavirus disease 2019 (COVID-19) appeared in Wuhan (Hubei, China) and subsequently swept the globe. In addition to the risk of infection, there is a strong possibility that post-traumatic stress disorder (PTSD) may be a secondary effect of the pandemic. Health care workers (HCWs) participating in the pandemic are highly exposed to and may bear the brunt out of stressful or traumatic events. In this cross-sectional study, we assessed the morbidity and risk factors of PTSD symptoms among Chinese HCWs. A total of 457 HCWs were recruited from March 15, 2020, to Mach 22, 2020, including HCWs in Wuhan and Hubei Province (excluding Wuhan), the areas first and most seriously impacted by COVID-19. The morbidity of PTSD symptoms was assessed by the Event Scale–Revised (IES-R). The risk factors for PTSD symptoms were explored by means of logistic regression analysis. Over 40% of the respondents experienced PTSD symptoms more than one month after the COVID-19 outbreak, and this proportion increased to 57.7% in Wuhan HCWs, especially females and HCWs on the frontline. Thus, rapid mental health assessment and effective psychological interventions need to be developed for frontline HCWs to prevent long-term PTSD-related disabilities. Moreover, Negative coping style and neuroticism personality may be regarded as high risk factors for PTSD symptoms. Improving individual coping strategies to enhance resilience should be the focus of further preventive intervention strategies.

ABSTRACT

Objective: To explore the value of liver function indexes on evaluation of the illness condition of coronavirus disease 2019 (COVID-19). Methods: 261 patients with confirmed COVID-19 which collected from Huangshi Hospital of Traditional Chinese Medicine from January to March 2020 were investigated and separated into:group of critical type, group of severe type and group of common type, and the data of the patients about age, gender, past medical history and the results of liver function test were collected. Chi-square test, analysis of variance, univariate and multivariate logistic regression analysis were adopted to explore the relationship between liver function indexes and illness condition of COVID-19. Results: 50.2% of COVID-19 patients had abnormal liver function. Compared with the group of severe type, the levels of serum alanine aminotransferase (ALT), aspartate aminotransferase(AST), alkaline phosphatase (ALP), P-glutamyl transferase (GGT) and total bilirubin (TBIL)in the group of critical type was significantly higher, while the level of albumin(ALB)was significantly lower, and the differences were statistically significant (all P < 0.05);compared with the group of common type, the levels of ALT, +AST, and GGT in the group of severe type were significantly higher, while the level of ALB was significantly lower, and the differences were statistically significant (all P < 0.05). The proportions of patients with abnormal liver function or liver damage in the group of critical type were significantly higher than those in the group of severe type (P < 0.05), and the proportions of patients with abnormal liver function or liver damage in the group of severe type was significantly higher than those in the group of common type (P < 0.05). The incidence ratio of abnormal liver function in patients with underlying disease was higher than that of without underlying disease (P < 0.05). ALT, AST, ALP, TBIL, and ALB were all risk factors for severe progress of COVID-19 disease (all P < 0.05);multivariate logistic regression analysis inidicating that TBIL (OR=10.862, P < 0.05) and ALB (OR=11.733, P < 0.05)were the independent risk factors. TBIL level was positively correlated with the severity of COVID-19 (r=0.367, P < 0.05), and ALB level was negatively correlated with the severity of COVID-19 (r=-0.613, P < 0.05). Conclusions: The abnormal liver function, especially the obvious abnormality of TBIL and ALB, could be used as the reference index of COVID-19 severity. The COVID-19 patients with underlying disease were easily suffered liver injury.

ABSTRACT

The pandemic of COVID-19 is now posing a worldwide hazard to one's health. Exposure to COVID-19 may cause negative emotions like anxiety, which is one of recognized risk factors for aggressive behaviors. This study looked into the effect of exposure to COVID-19 on aggression and how anxiety may act as a mediating factor, as well as lastly how rumination could moderate a variety of indirect paths during the epidemic of COVID-19. According to the current study's findings, which included a sizable sample of Chinese college students (N = 1,518), being exposed to COVID-19 showed a positive connection with aggression and anxiety, as well as rumination. These findings clarify the role that mediators play in the relationship between anxiety and exposure to COVID-19. The results are also helpful for personalizing treatments and putting preventative measures in place to decrease the aggression brought on by exposure to COVID-19. It is explored how lowering rumination and anxiety may be useful in the context of COVID-19 to lessen the psychopathological effects of the condition.

ABSTRACT

Big data technology plays an important role in the prevention and control of public health emergencies such as the COVID-19 pandemic. Current studies on model construction, such as SIR infectious disease model, 4R crisis management model, etc., have put forward decision-making suggestions from different perspectives, which also provide a reference basis for the research in this paper. This paper conducts an exploratory study on the construction of a big data prevention and control model for public health emergencies by using the grounded theory, a qualitative research method, with literature, policies, and regulations as research samples, and makes a grounded analysis through three-level coding and saturation test. Main results are as follows: (1) The three elements of data layer, subject layer, and application layer play a prominent role in the digital prevention and control practice of epidemic in China and constitute the basic framework of the "DSA" model. (2) The "DSA" model integrates cross-industry, cross-region, and cross-domain epidemic data into one system framework, effectively solving the disadvantages of fragmentation caused by "information island". (3) The "DSA" model analyzes the differences in information needs of different subjects during an outbreak and summarizes several collaborative approaches to promote resource sharing and cooperative governance. (4) The "DSA" model analyzes the specific application scenarios of big data technology in different stages of epidemic development, effectively responding to the disconnection between current technological development and realistic needs.

ABSTRACT

In order to explore the hydrochemical characteristics, influencing factors, and water quality of various water bodies in Bangong Co Lake Watershed, 60 water samples were collected from lake, river, groundwater, glacier water bodies in the watershed. Piper diagram, Gibbs’ diagrams, ion ratio analysis, statistical methods, and principal component analysis were used to study the hydrochemical characteristics and its influencing factors. Drinking water quality index (DWQI) and USSL classification were applied to assess the groundwater quality suitability for agricultural and drinking purposes. The hydrochemical characteristics show the differences among water bodies and their spatial distribution. Analyzed groundwater and surface water samples such as river water and glaciers mainly presented Ca-HCO3 type, and lake water mainly presented Na-Cl type and a small number of Na-HCO3·Cl type. The lake water chemical components are mainly affected by evaporative karst decomposition. The main mineralization process of groundwater and river water was related to the dissolution of reservoir minerals such as dolomite and calcite, and halite. The DWQI indicates that 79% of the groundwater samples in the study area showed a good quality for drinking. For irrigation water quality, the electrical conductivity (EC), calculated Sodium adsorption ratio (SAR), Magnesium hazardous ratio (MHR) showed that more than 13% of the total samples were not suitable for irrigation. USSL classification indicated that glacier and river water are relatively suitable for irrigation. And part of the groundwater and lake water has very high alkalinity or salinity which is alarming when considered for irrigation.

The consumption of famciclovir (FCV) has been increased dramatically since the outbreak of coronavirus in 2019, and the pollution and harm of FCV in waters are concerned. Here, by utilizing aryl halides on 2, 4, 6-tris(4-bromophenyl)- 1, 3, 5-triazine (BPT) and primary amine groups on benzidine (BZ), a novel conjugated microporous polymer, namely BPT-BZ-CMP, was synthesized by Buchwald-Hartwig coupling reaction and applied in the removal of FCV from aqueous solution firstly. The synthesized BPT-BZ-CMP were characterized by various methods, including FTIR, SEM, BET, and Zeta-potential. Due to the micropore structure and high specific surface area, it took only 30 min for BPT-BZ-CMP to adsorb FCV to reach an equilibrium, and the maximum adsorption capacity was 347.8 mg·g-1. The Liu and pseudo-second-order kinetic models properly fit the adsorption equilibrium and kinetic data, respectively. The adsorption process was a spontaneous process, and the hydrogen bonding, π-π interaction and C-H···π interaction enhanced the adsorption of FCV on BPT-BZ-CMP. BPT-BZ-CMP maintained a good adsorption capacity after four consecutive adsorption-desorption cycle experiments. This study confirmed the potential of BPT-BZ-CMP as efficient sorbent to remove FCV from aqueous solutions.

The global outbreak of COVID-19 has emerged as one of the most devastating and challenging threats to humanity. As many frontline workers are fighting against this disease, researchers are struggling to obtain a better understanding of the pathways and challenges of this pandemic. This paper evaluates the concept that the transmission of COVID-19 is intrinsically linked to temperature. Some complex nonlinear functional forms, such as the cubic function, are introduced to the empirical models to understand the interaction between temperature and the "growth" in the number of infected cases. An accurate quantitative interaction between temperature and the confirmed COVID-19 cases is obtained as: log(Y) = -0.000146(temp_H)3 + 0.007410(temp_H)2 - 0.063332 temp_H + 7.793842, where Y is the periodic growth in confirmed COVID-19 cases, and temp_H is the maximum daily temperature. This equation alone may be the first confirmed way to measure the quantitative interaction between temperature and human transmission of COVID-19. In addition, four important regions are identified in terms of maximum daily temperature (in Celsius) to understand the dynamics in the transmission of COVID-19 related to temperature. First, transmission decreases within the range of -50°C to 5.02°C. Second, the transmission accelerates in the range of 5.02°C to 16.92°C. Essentially, this is the temperature range for an outbreak. Third, transmission increases more slowly in the range of 16.92°C to 28.82°C. Within this range, the number of infections continue to grow, but at a slower pace. Finally, transmission decreases in the range of 28.82°C to 50°C. Thus, according to this hypothesis, the threshold of 16.92°C is the most critical, as the point at which infection rate is the greatest. This result sheds light on the mechanism in the cyclicity of the ongoing COVID-19 pandemic worldwide. The implications of these results on policy issues are also discussed in relation to a possible cyclical fluctuation pattern between the Northern and Southern Hemispheres.

ABSTRACT

Cytokine storm is a phenomenon whereby the overreaction of the human immune system leads to the release of inflammatory cytokines, which can lead to multiple organ dysfunction syndrome. At present, the existing drugs for the treatment of cytokine storm have limited efficacy and severe adverse effects. Here, we report a lymphatic targeting self-microemulsifying drug delivery system containing baicalein to effectively inhibit cytokine storm. Baicalein self-microemulsion with phospholipid complex as an intermediate carrier (BAPC-SME) prepared in this study could be spontaneously emulsified to form 12-nm oil-in-water nanoemulsion after administration. And then BAPC-SME underwent uptake by enterocyte through endocytosis mediated by lipid valve and clathrin, and had obvious characteristics of mesenteric lymph node targeting distribution. Oral administration of BAPC-SME could significantly inhibit the increase in plasma levels of 14 cytokines: TNF-α, IL-6, IFN-γ, MCP-1, IL-17A, IL-27, IL-1α, GM-CSF, MIG, IFN-ß, IL-12, MIP-3α, IL-23, and RANTES in mice experiencing systemic cytokine storm. BAPC-SME could also significantly improve the pathological injury and inflammatory cell infiltration of lung tissue in mice experiencing local cytokine storm. This study does not only provide a new lymphatic targeted drug delivery strategy for the treatment of cytokine storm but also has great practical significance for the clinical development of baicalein self-microemulsion therapies for cytokine storm.

ABSTRACT

The 3C-like protease (3CLpro) is essential for the replication and transcription of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), making it a promising target for the treatment of corona virus disease 2019 (COVID-19). In this study, a series of 2,3,5-substituted [1,2,4]-thiadiazole analogs were discovered to be able to inhibit 3CLpro as non-peptidomimetic covalent binders at submicromolar levels, with IC50 values ranging from 0.118 to 0.582 µM. Interestingly, these compounds were also shown to inhibit PLpro with the same level of IC50 values, but had negligible effect on proteases such as chymotrypsin, cathepsin B, and cathepsin L. Subsequently, the antiviral abilities of these compounds were evaluated in cell-based assays, and compound 6g showed potent antiviral activity with an EC50 value of 7.249 µM. It was proposed that these compounds covalently bind to the catalytic cysteine 145 via a ring-opening metathesis reaction mechanism. To understand this covalent-binding reaction, we chose compound 6a, one of the identified hit compounds, as a representative to investigate the reaction mechanism in detail by combing several computational predictions and experimental validation. The process of ring-opening metathesis was theoretically studied using quantum chemistry calculations according to the transition state theory. Our study revealed that the 2,3,5-substituted [1,2,4]-thiadiazole group could covalently modify the catalytic cysteine in the binding pocket of 3CLpro as a potential warhead. Moreover, 6a was a known GPCR modulator, and our study is also a successful computational method-based drug-repurposing study.

Novel coronavirus disease 2019 (COVID-19) has rapidly spread throughout the world; however, it is difficult for clinicians to make early diagnoses. This study is to evaluate the feasibility of using deep learning (DL) models to identify asymptomatic COVID-19 patients based on chest CT images. In this retrospective study, six DL models (Xception, NASNet, ResNet, EfficientNet, ViT, and Swin), based on convolutional neural networks (CNNs) or transformer architectures, were trained to identify asymptomatic patients with COVID-19 on chest CT images. Data from Yangzhou were randomly split into a training set (n = 2140) and an internal-validation set (n = 360). Data from Suzhou was the external-test set (n = 200). Model performance was assessed by the metrics accuracy, recall, and specificity and was compared with the assessments of two radiologists. A total of 2700 chest CT images were collected in this study. In the validation dataset, the Swin model achieved the highest accuracy of 0.994, followed by the EfficientNet model (0.954). The recall and the precision of the Swin model were 0.989 and 1.000, respectively. In the test dataset, the Swin model was still the best and achieved the highest accuracy (0.980). All the DL models performed remarkably better than the two experts. Last, the time on the test set diagnosis spent by two experts-42 min, 17 s (junior); and 29 min, 43 s (senior)-was significantly higher than those of the DL models (all below 2 min). This study evaluated the feasibility of multiple DL models in distinguishing asymptomatic patients with COVID-19 from healthy subjects on chest CT images. It found that a transformer-based model, the Swin model, performed best.

Consumerism during the COVID-19 pandemic has been characterized by impulsive buying. Using the theoretical lens of uncertainty avoidance and ego-depletion to identify the mediating mechanisms and moderating factors for online impulse buying, we surveyed young consumers across two relevant periods for high consumerism—the week preceding the 2021 Chinese Spring Festival (Study 1;February 4–9, 2021, n = 1495) and the weeks during and after the festival (Study 2;February 12 to March 2, 2021, n = 923). Perception of COVID-19 variant uncertainty was both directly and indirectly (via online shopping trust) positively associated with online impulse buying. COVID-19 burnout was consistently indirectly associated with online impulse buying via self-regulation and self-appraised impulsivity but inconsistently directly associated. Self-regulation was surprisingly positively associated with online impulse buying, possibly reflecting evidence of already depleted resources from prolonged regulatory exertion among high self-regulators. Self-appraised impulsivity negatively interacted with perception of COVID-19 variant uncertainty, suggesting that as trait impulsivity increases, individuals are less incentivized by peripheral drivers of online impulse buying.

ABSTRACT

Background: Coronavirus disease 2019 (COVID-19) have brought great disaster to mankind, and there is currently no globally recognized specific drug or treatment. Severe COVID-19 may trigger a cytokine storm, manifested by increased levels of cytokines including interleukin-17 (IL-17), so a new strategy to treat COVID-19 may be to use existing IL-17 inhibitors, which have demonstrated efficacy, safety and tolerability in the treatment of psoriasis. However, the use of IL-17 inhibitors in patients with psoriasis during the COVID-19 pandemic remains controversial due to reports that IL-17 inhibitors may increase the risk of respiratory tract infections. Objectives: The systematic review and meta-analysis aimed to evaluate the effect of IL-17 inhibitors on the risk of COVID-19 infection, hospitalization, and mortality in patients with psoriasis. Methods: Databases (including Embase, PubMed, SCI-Web of Science, Scopus, CNKI, and the Cochrane Library) were searched up to August 23, 2022, for studies exploring differences in COVID-19 outcomes between psoriasis patients using IL-17 inhibitors and those using non-biologics. Two authors independently extracted data and assessed the risk of bias in a double-blind manner. The risk ratios (RRs) and 95% confidence intervals (CIs) were calculated and heterogeneities were determined by the Q test and I 2 statistic. And the numbers needed to treat (NNTs) were calculated to assess the clinical value of IL-17 inhibitors in preventing SARS-CoV-2 infection and treating COVID-19. Results: Nine observational studies involving 7,106 participants were included. The pooled effect showed no significant differences in the rates of SARS-CoV-2 infection (P = 0.94; I 2 = 19.5%), COVID-19 hospitalization (P = 0.64; I 2 = 0.0%), and COVID-19 mortality (P = 0.32; I 2 = 0.0%) in psoriasis patients using IL-17 inhibitors compared with using non-biologics. Subgroup analyses grouped by age and COVID-19 cases, respectively, revealed consistent results as above. Meanwhile, the pooled NNTs showed no significant differences between the two groups in the clinical value of preventing SARS-CoV-2 infection and treating COVID-19. Conclusion: The use of IL-17 inhibitors in patients with psoriasis does not increase the risk of SARS-CoV-2 infection or worsen the course of COVID-19. ABSTRACT

BACKGROUND: From the end of 2019 to now, coronavirus disease 2019 (COVID-19) has put enormous strain on the world's health systems. As a characteristic sign of COVID-19 patient, olfactory dysfunction (OD) poses considerable problems for patients. In China, acupuncture has been widely used to treat OD caused by COVID-19, but there is still a lack of evidence-based medical evaluation. This study was designed to evaluate the effectiveness and safety of acupuncture for the treatment of COVID-19 OD. METHODS: According to the retrieval strategies, randomized controlled trials on the acupuncture for COVID-19 OD were obtained from Cochrane Central Register of Controlled Trials, Embase, PubMed, Web of Science, the Chinese National Knowledge Infrastructure, the Chinese Biomedical Literature Database, the Chinese Scientific Journal Database and the Wanfang Database, regardless of publication date, or language. Studies were screened based on inclusion and exclusion criteria, and the Cochrane risk bias assessment tool was used to evaluate the quality of the studies. The meta-analysis was performed using Review Manager (RevMan 5.3) and STATA 14.2 software. Ultimately, the evidentiary grade for the results will be evaluated. RESULTS: The results of this meta-analysis will be submitted to a peer-reviewed journal for publication. CONCLUSION: This study will provide up-to-date summary proof for evaluating the effectiveness and safety of acupuncture for COVID-19 OD.
